The CelVivo system creates an environment which promotes the growth and maintenance of large 3D tissue mimetic structures, whether they are spheroids, organoids and other aggregates. The technology CelVivo has developed to grow cells so they retain or recover their in vivo physiological attributes.

Application in Focus: Cancer Research

When it comes to replicating in vivo microenvironments, it is clear that 3D culture is far superior to 2D. To generate 3D constructs which will mostly closely mimic in vivo conditions, it is necessary to culture them in an environment that is an accurate biological representation. The ClinoStar system is a 3D bioreactor platform which employs the clinostat principle to grow spheroids. It provides a ‘microgravity’ simulated environment which produces highly reproducible 3D cultures. The system enables cultures to be grown and mature over longer periods of time, thus facilitating the investigation of mimetic tissues that is comparable to physiological conditions.

Understanding the CelVivoTechnology

CelVivo’s ambition is to provide cells with conditions which allow them to develop functionality which closely mimics that seen in the intact organism.

To achieve this, they have developed a system which obviates the need for scaffolds, gels or any type of additives which might perturb gene expression away from its natural baseline equilibrium. Furthermore they have very significantly reduced contact with plastics and reduced shear forces. It is possible to use the culture media that you normally use for cell culture. CelVivo suggests reducing glucose levels to 1 g/L glucosw, which is more relative to those seen in healthy animals.

The CelVivo System components

  • Clinostar™ System,
  • Clinoreactors
  • Tablet to control the system.

The ClinoStar is a clinostat for 3D cell culture incorporated in a CO₂ incubator . Allowing you to culture cells in 3D in a single unit.

A tablet is used to control the temperature, CO₂ level and each of the 6 clinostat motors are (independently) controlled using an intuitive software. The CO₂ and temperature in the chamber is blended by a centrally located fan to ensure a precise, homogeneous growth environment.

Learn how the Clinostar principle works

The video explains how spheroids from immortal cell lines are formed in the Clinnoreactor. The spheroids will mature in the Clinoreactor over 18 days. After maturation, the spheoirds will exhibit functionality and architecture like in vivo tissue, making them the perfect model for studies in toxicology, efficacy and cell development.

The Advantages of using Clinostats for culturing cells

  • Large spheroids are created with high uniformity in size
  • Excellent for co-cultures and tumor studies
  • The cells experience very low shear stress
  • Active diffusion which allow good nutrient exchange.
  • The function, architecture and ultrastructure of the spheroid mimics that see in vivo after 3 weeks

Applications and Research Areas

The technology created by CelVivo allows generation of uniform, reproducible and functional spheroids and organoids. These spheroids mimic the function, structure and architecture of in vivo cells. Our technology isn’t merely “3D cell culture”, it lets customers perform research using cell models with an unprecedented correlation between in vivo and in vitro conditions, and closes the gap between 2D cell culture and cell function.
